Baclofen is capable of inhibiting both monosynaptic and polysynaptic reflexes at the spinal level, possibly by hyperpolarization of afferent terminals, although actions at supraspinal sites may also occur and contribute to its clinical effect.Although Baclofen is an analog of the putative inhibitory neurotransmitter gamma-aminobutyric acid (GABA), there is no conclusive evidence that actions on GABA systems are involved in the production of its clinical effects.

Tizanidine is a central alpha-2-adrenergic receptor agonist and presumably reduces spasticity by increasing presynaptic inhibition of motor neurons. The effects of Tizanidine are greatest on polysynaptic pathways. The overall effect of these actions is thought to reduce facilitation of spinal motor neurons.

Baclofen:GABAB-Agonist, 

經由Gi protein去打開神經末梢的鉀離子通道, 造成鉀離子往外跑的變多，產生過極化；

讓鈣離子通道不易打開，使鈣離子跑進來的變少，讓Glutamate的釋放也變少了，然後就會抑制突觸性反射與脊髓通路的傳導。

所以不是直接抑制或接觸骨骼肌，是間接抑制使骨骼肌收縮

作為骨骼肌鬆弛劑

治肌肉疼痛和痙攣


Diazepam:屬BZD類

(字尾-azpam中,

az=benzodiazepine,

p=positive,

a=allosteric,

m=modulator)

字尾三個字直接跟大家講機轉是GABAA的正相異位調節者

(非GABAA-Agonist),此藥會跟GABAA受體上以異位方式結合而增加GABA的親和力使GABAA打開Cl-通道的頻率變高。
