(2)增加尿酸排除藥物,如benzbromarone、sulfinpyrazone及probenecid; 促進尿酸排除藥物皆是弱有機酸,與尿酸競爭近端腎小管的尿酸-氫離子通道,減少尿酸重新吸收至體內的機會。這類藥品需注意可能產生尿路結石的情況,因此服用藥物期間,需注意飲水、減少尿路結石的情況發生。另外,這些藥品在腎功能不佳時,如probenecid在Ccr<50 ml/min、benzbromarone在Ccr<20 ml/min時,應避免使用。常見副作用為腸胃不適、腹瀉或尿路結石;常見交互作用為probenecid可能增加penicillin類的抗生素的濃度。 |

Lesch–Nyhan syndrome (LNS) is a rare inherited disorder caused by a deficiency of the enzyme hypoxanthine-guanine phosphoribosyltransferase (HGPRT). This deficiency occurs due to mutations in the HPRT1 gene located on the X chromosome. LNS affects about 1 in 380,000 live births The HGPRT deficiency causes a build-up of uric acid in all body fluids. The combination of increased synthesis and decreased utilization of purines leads to high levels of uric acid production.
