Take a / the day off 請一天假
Take a day off 或是 take the day off 都是請一天假的意思，唯一的差別是，take a day off 單純只是說要請一天的假，但還不確定是哪一天，而 take the day off 是已經確定要請某一天假時可以用，通常會指當天。

補充：
1.Be on leave / go on leave 請假
Leave 通常是指自己另外申請的假，如病假（sick leave）、生理假（menstruation leave）或是年假（annual leave）。
Ex.
Cathy is on leave this Thursday.
Cathy 這週四請假。

2.Take a sick leave 請病假
