A basic amine is an essential feature of all opioids because, in cationic conjugate acid form, it forms an essential electrostatic bond with the Asp residue conserved in all GPCRs.

ㆍWith the exception of dezocine, an opioid agonist no longer available in the U.S., only tertiary amines provide clinically significant central opioid agonist or antagonist activity.
